ABSTRACT

This experimental study was aimed to evaluate the injurious effects of chronic traumatic brain injury on cortex mitochondrial function in rats. The head of rat was impacted by a metal sphere in a weight-drop device twice per day for 30 days, cortex mitochondria were isolated. Then the mitochondria membrane fluidity, swelling, respiratory function, the activities of mitochondria respiratory enzymes and superoxide dismutase (SOD), the levels of phospholipid, malondial dehyde (MDA) and Ca2+ were determined to analyze the function of mitochondria. The data indicated that chronic closed traumatic brain injury caused severe neuronal mitochondrial injuries. The swelling of mitochondria was aggravated, the decomposability of mitochondrial membrane phospholipid was increased, the membrane fluidity of mitochondria was decreased; the chronic closed traumatic brain injury also significantly depressed the activities of respiratory enzymes and SOD of mitochondria, increased the level of MDA and Ca2+. The chronic closed traumatic brain injury induced damage to rat cortex mitochondria. The mechanisms may be derived from the secondary increase of free radicals induced by mitochondrial membrane injury and the obstacle of rat brain energy metabolism.

ABSTRACT

AIM To study the effects of polydatin on free radical induced rat cortex mitochondria injury. METHODS Fe 2++VitC system was used to produce ?OH. The mitochondria was isolated. Mitochondria membrane fluidity, swelling and contents of phospholipid were determined to measure the function of mitochondria membrane. The activities of ATPase and Cytochrome C oxidase were determined to measure the ability of mitochondria energy metabolism. The activity of superoxide dismutase (SOD) and content of malondial dehyde (MDA) were determined to measure the ability of anti oxygenation. RESULTS ?OH resulted in severe neuronal mitochondria injuries and the injuries and the injuries was alleviated by Polydatin (content of 100,200,400 mg?L -1). The swelling of mitochondria was ameliorated, the decomposability of mitochondrion membrane phospholipid was decreased, the membrane fluidity of mitochondria was increased. Polydatin also significantly inhibited the decrease in SOD, Cytochrome C oxidase and ATPase activity and the increase in MDA levels caused by free radical. CONCLUSION Polydatin has a protective action against the rat neuronal mitochondria injuries induced by oxygen free radical. The mechanisms may be derived from scavenging free radicals, reducing lipid peroxides, and improving the energy metabolism.

ABSTRACT

Object To set up a determination method for aucubin in three medicinal plants of Pedicularis L Methods The medicinal herbs were defatted by Et 2O, then extracted by MeOH and determined by HPLC method. Varian C 18 column, mobile phase: MeOH H 2O (15∶85); detection wavelength: 204 nm, flow rate: 0.5 mL/min. Results The content of aucubin and its recovery in three medicinal plants were as follows: aucubin in P. decora, in P. davidii and P. artselaeri were 0.90%, 1.94%, 0.95%; recovery rates were 102.3%, 98.2%, 97.6% in P. artelaeri. Conclusion Medicinal plants of Pedicularis Linn. has a good exploiting prospect, the research can provide the reference for evaluating of these medicinal herbs

ABSTRACT

AIM: To evaluate the antioxidant properties in vitro of Longxuejie Capsule (Sanguis draconis) (LC). METHODS: The antioxidant activities of Longxuejie Capsules were investigated by employing several external assay systems, including 1,1-diphenyl-2-picrylhydrazyl (DPPH?)/peroxide anion/hydroxyl radical-scavenging, reducing power assay system, peroxidation of polyunsaturated fatty acid from yolk lipoprotein induced by Fe~ 2+ method, and ?-carotene/linoleic acid assay system. RESULTS: The LC showed scavenging activity against free radicals, such as DPPH?, peroxide anion and hydroxyl radicals. LC also exhibited effective reducing power and powerful inhibitory effects on peroxidation of polyunsaturated fatty acid from yolk lipoprotein induced by Fe~ 2+ . Furthermore, LC showed markedly inhibitory effect on ?-carotene/linoleic acid assay system. CONCLUSION: Longxuejie Capsule has obvious antioxidant activities in vitro

ABSTRACT

Corn meal was hydrolyzed in HCI ,and pH adjusted to 6.8 with (NH_4)OH. The substrate was inoculated with Cadida trapicalis ,then cultivated in a shaker at 30℃ and in 200r/min for 2 days. The Results indicated that the protein content in corn meal grew from 8.4 to 27.4%the lysine in the protein increased from 32.1 to 73.48/kg,the tryptophan from 6.8 to to 14.8g/kg. This shows that the nutritional value of corn meal can be improved with this method.

ABSTRACT

Objective: To study the anti oxidative effects of PLF. Methods: The ?OH scavenging effect was measured by salicylic acid method, the content of MDA in liver mitochondria and microsome was measured by TBA assay; hemolysis of RBC and the swelling of mitochondria were detected by spectrophotometric methods. Results: PLF can scavenge ?OH, inhibit production of MDA, swelling of mitochondria and hemolysis of RBC. Conclusion: PLF has anti oxidation effects invitro.

ABSTRACT

Objective: To study the anti-senile and anti-oxidative activities of Forsythia suspense leaves tea(FSLT) in mice. Methods: 36 ICR mice(7 mo age) were randomly divided into three groups: senior control group,ig distilled water ; FSLT1 group,ig FSLT 0.3 g ?kg-1?d-1 ; and FSLT2 group,ig FSLT 0.9 g?kg-1?d-1 .The other 12 ICR mice(young control group) were given ig distilled water. MDA and MAO-B in brain and mitochondria were measured. SOD,MDA and POD in heart, liver index, thymus index and spleen index were determined. Results: Forsythia suspense leaves tea may reduce the level of MDA and MAO-B, and increase the activities of SOD and POD. Conclusion: Forsythia suspense leaves tea may have anti-senile and anti-oxidative activities.
